Porcine Parathyroid Hormone ELISA Kit

From Antibodies.com, this competitive Enzyme-Linked Immunosorbent Assay (cELISA) enables in vitro quantitative determination of porcine Parathyroid Hormone in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses a pre-coated 12 x 8 well strip microplate, is read on a colorimetric microplate reader at 450 nm, and has a total run time of 2h 30m.

The quantitative measuring range is 15.6–1000 pg/ml with a sensitivity of <5.86 pg/ml. In the competitive enzyme immunoassay format, Parathyroid Hormone present in samples or standards competes with pre-coated antigen for binding sites on a Biotinylated Anti-Parathyroid Hormone Antibody. After washing, an HRP-Avidin conjugate is added, followed by TMB substrate, which produces a yellow product upon addition of acidic stop solution; the resulting colour intensity is inversely proportional to the analyte concentration.

Validation across serum, EDTA plasma and heparin plasma yielded average recoveries of 91% (range 80–102%), 90% (range 81–99%) and 84% (range 80–89%), respectively (n=5). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 spanned 74% to 108% across the three sample types. Kit components are stored at +4°C or -20°C as instructed, and the product must not be used past its expiration date.
