Porcine Osteopontin ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) supplied by Antibodies.com for the in vitro quantitative determination of porcine Osteopontin. The kit is reactive against porcine samples and is intended for use with serum, plasma, tissue homogenates, cell lysates, or other biological fluids.

The assay uses a pre-coated microplate in a 12 x 8 well strip format. An antibody specific for Osteopontin is immobilised on the plate; standards and test samples are added and any Osteopontin present is captured by the immobilised antibody. After washing, a Biotinylated Anti-Osteopontin Antibody is added, followed by an HRP-Avidin conjugate and TMB substrate. The intensity of the yellow colour developed after addition of acidic stop solution is read at 450 nm and is proportional to Osteopontin concentration, which is calculated against the standard curve. The measuring range is 62.5–4000 pg/ml with a sensitivity of 29.8 pg/ml, and the assay is read on a colorimetric microplate reader. Total assay time is 4h 30m.

Reported recovery values are: serum (n=5) 80–102%, average 91%; EDTA plasma (n=5) 81–99%, average 90%; heparin plasma (n=5) 80–89%, average 84%. Linearity data across 1:2, 1:4, 1:8, and 1:16 dilutions is provided for serum, EDTA plasma, and heparin plasma. Kit components are stored at +4°C or –20°C as instructed and the kit must not be used past its expiration date.
