Porcine Osteocalcin ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) kit from Antibodies.com is designed for the in vitro quantitative determination of porcine Osteocalcin in serum, plasma, tissue homogenates and other biological fluids. The assay employs the sandwich enzyme immunoassay technique using a pre-coated 96-well microtiter plate. An antibody specific for Osteocalcin is immobilised on the plate; standards and test samples are added so that Osteocalcin in each sample is bound, followed by washing and incubation with a Biotinylated Anti-Osteocalcin Antibody. HRP-Avidin conjugate is then added, and TMB substrate solution is used to visualise the reaction, producing a blue-coloured product that turns yellow after addition of an acidic stop solution. Absorbance is read at 450 nm in a microplate reader and the Osteocalcin concentration is calculated from the standard curve.

Quantitative performance includes a sensitivity of <0.25 ng/ml and a working range of 0.781–50 ng/ml. The assay is read on a colorimetric microplate reader, with a total run time of 4 hours 30 minutes. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date. Reported recovery performance covers serum, EDTA plasma and heparin plasma in both neat and serially diluted formats.
