The kit uses the sandwich enzyme immunoassay technique, with an antibody specific for NT-ProBNP pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Standards and samples are added to the wells, followed by a Biotinylated Anti-NT-ProBNP Antibody and an HRP-Avidin conjugate. TMB substrate is added to visualise the enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of the acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader and the NT-ProBNP concentration is calculated by reference to the standard curve.

The assay has a reported sensitivity of <6.93 pg/ml and a working range of 15.6-1000 pg/ml, with a total run time of approximately 4 hours 30 minutes. Recovery has been evaluated in serum (80%-102%), EDTA plasma (81%-101%) and heparin plasma (80%-89%) across dilution factors from 1:2 to 1:16.
