Description

This sandwich ELISA kit from Antibodies.com enables the in vitro quantitative determination of porcine Monocyte Chemotactic Protein 2 (MCP2) in serum, plasma or other biological fluids. The assay uses a pre-coated 96-well microplate onto which an antibody specific for MCP2 has been immobilised. Standards and test samples are added to the wells, and MCP2 present in each sample is bound by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-MCP2 Antibody is added to bind the captured analyte, after which an HRP-Avidin conjugate is introduced. After further incubation and washing, a TMB substrate solution is used to visualise the enzymatic reaction, producing a blue product that changes to yellow upon addition of the acidic stop solution. Absorbance is measured at 450 nm in a colorimetric microplate reader and MCP2 concentration is calculated by reference to the standard curve.

The assay has a sensitivity of less than 32 pg/ml and a quantitative range of 78.1–5000 pg/ml, with a total assay time of 4 hours 30 minutes. In spike-and-recovery evaluation with n=5 samples, recoveries in serum ranged from 80–102% (average 91%), in EDTA plasma from 81–100% (average 90%), and in heparin plasma from 80–89% (average 84%). Dilution linearity was also assessed at 1:2, 1:4, 1:8 and 1:16 dilutions across the same three sample types. Kit components should be stored at +4°C or –20°C as instructed, and the kit must not be used past its expiration date.
