Porcine Matrix Metalloproteinase 9 ELISA Kit

Distributed by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of porcine Matrix Metalloproteinase 9 (MMP9) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The kit uses a sandwich enzyme immunoassay technique in which an antibody specific for MMP9 is pre-coated onto a 96-well microtiter plate arranged as 12 x 8 well strips. Standards and samples are added to the wells and any MMP9 present binds to the immobilised antibody; after washing, a Biotinylated Anti-MMP9 Antibody binds the captured MMP9, followed by an HRP-Avidin conjugate. TMB substrate solution is added to visualise the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to MMP9 concentration, read as O.D. absorbance at 450 nm in a microplate reader against the standard curve.

Stated performance specifications include a product range of 1.56–100 ng/ml and a sensitivity of 0.56 ng/ml, with a total assay time of 4h 30m and colourimetric detection on a microplate reader. Reported sample recovery averages are 91% for serum, 90% for EDTA plasma and 84% for heparin plasma (each n=5). Dilution linearity data are provided across 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma.
