Description
From Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of porcine Leukemia Inhibitory Factor (LIF) in serum, plasma, tissue homogenates or other biological fluids. The assay employs a pre-coated microplate in a 12 x 8 well strip format with an antibody specific for LIF, using a biotinylated anti-LIF detection antibody and an HRP-avidin conjugate. TMB substrate is used to visualise the HRP reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to LIF captured in each well, and the concentration is calculated from the O.D. absorbance read at 450nm in a colorimetric microplate reader.
The assay has a reported sensitivity of <0.047 ng/ml and a quantitative range of 0.156-10 ng/ml, with a typical total assay time of 4h 30m. Spike-and-recovery was evaluated across sample types: serum (n=5) yielded 80%-102% with an average of 91%; EDTA plasma (n=5) yielded 81%-99% with an average of 90%; and heparin plasma (n=5) yielded 80%-89% with an average of 84%. Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16 across the same sample types, with serum at 87-91%, 87-107%, 74-101% and 92-97%; EDTA plasma at 90-105%, 84-101%, 90-101% and 79-108%; and heparin plasma at 84-95%, 92-105%, 82-105% and 89-91%. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
