A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) supplied by Antibodies.com, this kit enables the in vitro quantitative determination of porcine Leptin Receptor in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay has a detection range of 1.56-100 ng/ml and a sensitivity of 0.59 ng/ml, with a total run time of 4h 30m.

The workflow uses a pre-coated 96-well microplate arranged as 12 x 8 well strips. Standards and test samples are captured by an immobilized antibody specific for Leptin Receptor, followed by incubation with a Biotinylated Anti-Leptin Receptor Antibody and an HRP-Avidin conjugate. Bound complex is visualised using TMB substrate, with the reaction stopped by acidic stop solution and absorbance read at 450 nm on a microplate reader.

Spike-and-recovery performance (n=5) gave average recoveries of 91% for serum (range 80%-102%), 90% for EDTA plasma (range 81%-100%), and 84% for heparin plasma (range 80%-89%). Dilution linearity was assessed at 1:2, 1:4, 1:8, and 1:16 dilutions, with serum (n=5) ranges of 87-91%, 87-107%, 74-101%, and 92-97% respectively; EDTA plasma (n=5) at 90-105%, 84-101%, 90-101%, and 79-108%; and heparin plasma (n=5) at 84-95%, 92-105%, 82-105%, and 89-91%. Kit components should be stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.
