Porcine Leptin ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of porcine Leptin in serum, plasma, tissue homogenates or other biological fluids. The kit uses the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate arranged as 12 x 8 well strips.

Standards and samples are added to the wells, where Leptin is captured by an immobilised antibody. After washing, a Biotinylated Anti-Leptin Antibody binds the captured analyte, followed by addition of an HRP-Avidin conjugate. TMB substrate is then added to visualise the HRP enzymatic reaction, producing a blue product that turns yellow after the addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader, and Leptin concentration is calculated from the standard curve.

The assay uses a quantitative colourimetric detection format with a run time of 4 hours 30 minutes, an assay range of 0.312–20 ng/ml and a sensitivity of <0.124 ng/ml. Kit components are stored at +4°C or –20°C as instructed. In spike-and-recovery testing (n=5 per matrix), average recoveries were 91% in serum (range 80–102%), 90% in EDTA plasma (range 81–99%) and 84% in heparin plasma (range 80–89%).
