Porcine Interleukin 18 ELISA Kit

Antibodies.com supplies this sandwich ELISA kit for the in vitro quantitative determination of porcine Interleukin 18 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a 96-well microtiter plate pre-coated with an antibody specific for IL-18; standards and samples are added so the analyte is captured by the immobilised antibody, then the wells are washed and incubated with a Biotinylated Anti-IL-18 Antibody, followed by an HRP-Avidin conjugate. After further washing, TMB substrate solution is added to visualise the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution; absorbance is read at 450 nm in a microplate reader and IL-18 concentration is calculated by reference to the standard curve.

Performance characteristics include a product range of 15.6-1000 pg/ml and a sensitivity of <5.8 pg/ml, with the full assay completed in 4h 30m. Spike-and-recovery has been characterised at n=5 in serum (range 80%-102%, mean 91%), EDTA plasma (range 81%-100%, mean 90%) and heparin plasma (range 80%-89%, mean 84%), and dilution linearity at 1:2, 1:4, 1:8 and 1:16 has been reported across the same three sample types. Detection is colourimetric via the pre-coated microplate arranged as 12 x 8 well strips, read on a standard microplate reader.

Kit components supplied include a pre-coated 96-well microplate, lyophilised standard, Detection Solution A, Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers, with storage at +4°C or -20°C as instructed in the protocol manual.
