Description

Manufactured by Antibodies.com, this Porcine IL-10 ELISA Kit is a sandwich enzyme-linked immunosorbent assay (sELISA) designed for the in vitro quantitative determination of porcine IL-10 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. An antibody specific for IL-10 has been pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells, where IL-10 is bound by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-IL-10 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The absorbance is read at 450nm in a colorimetric microplate reader.

The assay has a quantitative detection range of 15.6-1000 pg/ml and a sensitivity of 6.5 pg/ml, with a total assay time of 4h 30m. Kit components, including pre-coated microplate strips, lyophilised standard, detection solutions, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, are stored at +4°C or -20°C as instructed. The kit is suitable for serum, plasma, cell lysates, tissue homogenates and cell culture supernates.

Recovery has been validated across sample types, with average values of 94% for serum (n=5), 95% for EDTA plasma (n=5) and 96% for heparin plasma (n=5), with individual recovery ranges of 87%-101%, 90%-100% and 91%-102% respectively. Dilution linearity has been demonstrated at 1:2, 1:4, 1:8 and 1:16 dilutions across serum (82-96%, 83-98%, 81-99%, 93-101%), EDTA plasma (88-101%, 86-95%, 90-102%, 80-93%) and heparin plasma (80-91%, 82-90%, 95-104%, 79-95%) samples (n=5).
