From Antibodies.com, this Porcine Interferon Gamma ELISA Kit is a sandwich enzyme-linked immunosorbent assay (sELISA) intended for the in vitro quantitative determination of porcine interferon gamma in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, and other biological fluids. The assay uses a 96-well microtiter plate pre-coated with an antibody specific for interferon gamma; standards and test samples are added to the wells, with bound analyte detected using a biotinylated anti-interferon gamma antibody and an HRP-avidin conjugate. The enzymatic reaction is visualised with a TMB substrate that produces a blue product, which turns yellow on addition of an acidic stop solution.

The quantitative measurement range is 15.6–1000 pg/ml with a sensitivity of 5.9 pg/ml. Absorbance is read at 450 nm using a colorimetric microplate reader, and interferon gamma concentrations are calculated against the standard curve. The total assay time is 4h 30m, and the platform is a pre-coated microplate arranged as 12 × 8-well strips.

Typical recovery values reported with the kit are 80–102% for serum (n=5), 81–100% for EDTA plasma (n=5), and 80–89% for heparin plasma (n=5). Linearity across 1:2, 1:4, 1:8, and 1:16 dilutions is 87–91%, 87–107%, 74–101%, and 92–97% for serum; 90–105%, 84–101%, 90–101%, and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105%, and 89–91% for heparin plasma. Components are stored at +4 °C or –20 °C as instructed, and the kit must not be used past its expiration date.
