This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com enables the in vitro quantitative determination of porcine HIF-1 alpha in serum, plasma, tissue homogenates or other biological fluids. A capture antibody specific for HIF-1 alpha is pre-coated onto a 96-well microplate arranged as 12 x 8 well strips; bound analyte is detected with a biotinylated anti-HIF-1 alpha antibody and an HRP-avidin conjugate, then visualised using TMB substrate, with absorbance read at 450 nm on a colorimetric microplate reader. Total assay time is 4 hours 30 minutes.
The manufacturer reports a detection sensitivity of <5.8 pg/ml and an assay range of 15.6-1000 pg/ml. Spike-and-recovery averages reported by the manufacturer are 91% for serum (range 80-102%, n=5), 90% for EDTA plasma (range 81-100%, n=5) and 84% for heparin plasma (range 80-89%, n=5). Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past the stated expiration date.
