Porcine Heat Shock 70kDa Protein 1A ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) is designed for the in vitro quantitative determination of porcine Heat Shock 70kDa Protein 1A (HSP70) in serum, plasma or other biological fluids. An antibody specific for HSP70 is pre-coated onto a microplate, with standards and test samples added to the wells. Following incubation and washing, a Biotinylated Anti-HSP70 Antibody is applied, followed by an HRP-Avidin conjugate and TMB substrate. The catalysed reaction produces a blue product that changes to yellow upon addition of acidic stop solution, with colour density proportional to HSP70 concentration read at 450 nm.

The assay range is 3.12-200 ng/ml with a sensitivity of <1.14 ng/ml, and the total assay time is 4h 30m. The pre-coated 96 well microplate is read on a colorimetric microplate reader, and HSP70 concentration is calculated by reference to a standard curve. Kit components include a lyophilized standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, stored at +4°C or -20°C as instructed.

Validation data for serum, EDTA plasma and heparin plasma report average recoveries of 91% (serum, n=5), 90% (EDTA plasma, n=5) and 84% (heparin plasma, n=5), with recoveries assessed at dilutions of 1:2, 1:4, 1:8 and 1:16.
