Manufactured for Antibodies.com, the Porcine Granzyme M ELISA Kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) intended for the in vitro quantitative determination of porcine Granzyme M in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, and other biological fluids. The assay is run on a pre-coated 96-well microplate arranged as 12 x 8 well strips and uses a biotinylated anti-Granzyme M detection antibody, an HRP-avidin conjugate, and TMB substrate; absorbance is read at 450 nm on a colorimetric microplate reader. Total assay time is 4h 30m.

The kit reports a working range of 15.6–1000 pg/ml and a sensitivity of <8.9 pg/ml. According to the manufacturer datasheet, spiked recovery testing gave the following: serum (n=5) average 91% (range 80%–102%); EDTA plasma (n=5) average 90% (81%–100%); heparin plasma (n=5) average 84% (80%–89%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 dilutions was reported as: serum (n=5) 87–91%, 87–107%, 74–101%, 92–97%; EDTA plasma (n=5) 90–105%, 84–101%, 90–101%, 79–108%; heparin plasma (n=5) 84–95%, 92–105%, 82–105%, 89–91%. Supplied components include a pre-coated 96-well microplate, lyophilized standard, Detection Solution A, Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution, and adhesive plate sealers. Components are stored at +4°C or -20°C as indicated and should not be used past the expiration date.
