Porcine Ghrelin ELISA Kit

Antibodies.com supplies a competitive Enzyme-Linked Immunosorbent Assay (cELISA) for the in vitro quantitative determination of porcine Ghrelin in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses a 96-well microtiter plate pre-coated with Ghrelin antigen, in which Ghrelin present in standards or samples competes with the immobilized antigen for binding sites on a Biotinylated Anti-Ghrelin Antibody. After washing, an HRP-Avidin conjugate is added, and the enzymatic reaction is visualised using TMB substrate, producing a blue colour that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader, with colour intensity inversely proportional to Ghrelin concentration.

The kit offers a measuring range of 0.156–10 ng/ml and a sensitivity of 0.043 ng/ml, with a total assay time of 2h 30m. Reported recoveries for undiluted samples average 91% for serum (range 80%–102%, n=5), 90% for EDTA plasma (range 81%–100%, n=5) and 84% for heparin plasma (range 80%–89%, n=5). Supplied components, stored at +4°C or -20°C as instructed, include a pre-coated 96-well microplate (12 x 8 well strips), lyophilized standard (2 vials), Detection Solution A (70 μl), Detection Solution B (120 µl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and 5 adhesive plate sealers.
