Porcine Galectin 4 ELISA Kit

This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of porcine Galectin 4 (GAL4) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay employs the sandwich enzyme immunoassay technique, with an antibody specific for GAL4 pre-coated onto a 96-well microplate. Standards and test samples are added to the wells and bound by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-GAL4 Antibody is added, which binds the captured GAL4. An HRP-Avidin conjugate is then applied before incubation with TMB substrate solution. The enzymatic reaction produces a blue-coloured product that turns yellow upon addition of acidic stop solution; the density of yellow is proportional to the amount of GAL4 captured, and concentration is calculated from the O.D. absorbance at 450 nm using a microplate reader.

The kit reports a product range of 0.312–20 ng/ml and a sensitivity of 0.074 ng/ml. Total assay time is 4 hours 30 minutes. Reported recovery values are: Serum (n=5) 80%–102%, average 91%; EDTA Plasma (n=5) 81%–100%, average 90%; Heparin Plasma (n=5) 80%–89%, average 84%. Linear dilution performance across 1:2, 1:4, 1:8 and 1:16 dilutions was: Serum (n=5) 87–91%, 87–107%, 74–101%, 92–97%; EDTA Plasma (n=5) 90–105%, 84–101%, 90–101%, 79–108%; Heparin Plasma (n=5) 84–95%, 92–105%, 82–105%, 89–91%.
