Porcine Fibroblast Growth Factor 21 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of porcine Fibroblast Growth Factor 21 (FGF21). The kit is suitable for use with serum, plasma, tissue homogenates, cell lysates, cell culture supernates, and other biological fluids, with detection performed on a colorimetric microplate reader measuring absorbance at 450 nm following TMB substrate development.

The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips. An antibody specific for FGF21 is immobilised on the plate; standards and samples are added and FGF21 present is captured by the immobilised antibody. A biotinylated anti-FGF21 detection antibody is then added, followed by HRP-Avidin conjugate. The resulting enzymatic reaction produces a blue product that changes to yellow upon addition of acidic stop solution, with the yellow intensity proportional to the amount of FGF21 captured in each well.

The kit has a stated product range of 31.2-2000 pg/ml and a sensitivity of 13.3 pg/ml, with a total assay time of approximately 4 hours 30 minutes. Validated sample recovery in neat matrix (n=5 per type) was 80-102% for serum (average 91%), 81-100% for EDTA plasma (average 90%) and 80-89% for heparin plasma (average 84%). Kit components are stored at +4°C or -20°C as instructed in the protocol, and the kit must not be used past its expiration date.
