Description

Antibodies.com offers a Porcine FGF1 ELISA Kit designed as a sandwich enzyme-linked immunosorbent assay (sELISA) for the in vitro quantitative determination of porcine Fibroblast Growth Factor 1, Acidic. The kit is reactive with porcine samples and is suitable for serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids.

The assay uses the sandwich enzyme immunoassay technique. An antibody specific for FGF1 is pre-coated onto a 96-well microtiter plate. Standards and samples are added and the FGF1 present binds to the wells via the immobilized antibody. After incubation and washing, a Biotinylated Anti-FGF1 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of FGF1 captured and is read at 450 nm on a colorimetric microplate reader.

The assay has a working range of 15.6–1000 pg/ml and a sensitivity of <6.6 pg/ml, with a total assay time of 4 hours 30 minutes. Kit components should be stored at +4°C or −20°C as instructed and the kit should not be used past its expiration date. Spike-and-recovery in samples (n=5) was: serum 85–100% (average 92%), EDTA plasma 85–102% (average 93%), and heparin plasma 87–99% (average 93%). Linearity upon dilution at 1:2, 1:4, 1:8 and 1:16 ranged from 79–104% across the three sample types.
