From Antibodies.com, this Porcine Factor Related Apoptosis Ligand (Fas Ligand) ELISA Kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of porcine Fas Ligand in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids.

The kit employs a pre-coated 96-well microplate with an antibody specific for Fas Ligand. Standards and samples are added to the wells, where Fas Ligand is captured by the immobilised antibody. After washing, a biotinylated anti-Fas Ligand detection antibody is added, followed by an HRP-Avidin conjugate and TMB substrate solution. The enzymatic reaction produces a blue colour that changes to yellow upon addition of acidic stop solution, with absorbance read at 450 nm on a colorimetric microplate reader.

The assay has a product range of 23.4-1500 pg/ml and a sensitivity of 5.2 pg/ml, with a total assay time of 4h 30m. Recovery in serum averaged 91% (range 80-102%), in EDTA plasma averaged 90% (range 81-99%), and in heparin plasma averaged 84% (range 80-89%). Kit components include the pre-coated microplate, lyophilised standard, detection solutions, wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers, stored at +4°C or -20°C as instructed.
