A quantitative sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro determination of porcine erythropoietin (EPO) in serum, plasma, or other biological fluids. The assay is performed on a pre-coated 96-well microplate arranged as 12 x 8 well strips, using an HRP-Avidin conjugate and TMB substrate for colorimetric detection measured at 450 nm.

The reported sensitivity is <0.065 ng/ml with an assay range of 0.156-10 ng/ml. Total assay time is approximately 4 hours and 30 minutes. Standards and test samples are incubated in the pre-coated wells, where EPO is captured by the immobilized antibody. After washing, a biotinylated anti-EPO antibody is added, followed by HRP-Avidin conjugate and TMB substrate. The enzymatic reaction produces a blue product that converts to yellow upon addition of stop solution, with the absorbance intensity proportional to the EPO concentration in each well.

Validation studies report recovery ranges of 80%-102% (average 91%) in serum, 81%-99% (average 90%) in EDTA plasma, and 80%-89% (average 84%) in heparin plasma (n=5 per sample type). Dilution linearity has been characterized at 1:2, 1:4, 1:8, and 1:16 ratios across these matrices. Kit components should be stored at +4°C or -20°C as instructed, and the product should not be used past its expiration date.
