A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of porcine Endothelin 1. The kit is reactive with porcine samples, falls within the Assays and Kits category, and is suitable for serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. It uses a pre-coated 96-well microplate (12 x 8 well strips) with colorimetric detection read at 450 nm, and kit components are stored at +4°C or -20°C as instructed.

Endothelin 1 in standards and samples is captured by an antibody pre-coated onto the wells. After washing, a biotinylated anti-Endothelin 1 detection antibody is added, followed by an HRP conjugate and TMB substrate. The enzymatic reaction produces a blue product that converts to yellow upon addition of acidic stop solution, and the absorbance at 450 nm is directly proportional to the Endothelin 1 concentration as read against a standard curve. The kit is reported as specific for Endothelin 1 with no reported cross-reactivity with other analogs or variants.

Performance specifications differ between the two manufacturer datasheets cited. One lists a product range of 12.5–800 pg/ml and a sensitivity of 4.3 pg/ml, with recoveries of 80–102% (average 91%) in serum, 81–100% (average 90%) in EDTA plasma, and 80–89% (average 84%) in heparin plasma (n=5). The other lists a product range of 1.25–80 pg/mL and a sensitivity of less than 0.625 pg/mL, with recoveries of 90–105% (average 93%) in serum, 85–101% (average 94%) in EDTA plasma, and 85–105% (average 95%) in heparin plasma (n=10). Reported assay times also differ, from 4 hours 30 minutes to 2 hours.
