Porcine Creatine Kinase, Muscle ELISA Kit

It employs the sandwich enzyme immunoassay technique on a 96-well microtiter plate pre-coated with an antibody specific for Creatine Kinase MM. Standards and samples are added to the wells where the analyte binds to the immobilized capture antibody, followed by incubation with a biotinylated anti-Creatine Kinase MM antibody and an HRP-avidin conjugate. The reaction is visualised with TMB substrate solution, producing a blue product that turns yellow on addition of acidic stop solution, and the absorbance is read at 450 nm in a colorimetric microplate reader. The reported product range is 9.38–600 U/L with a sensitivity of 3.96 U/L, and the assay can be completed in 4 h 30 min.

Spike-and-recovery was evaluated in serum (range 80%–102%, average 91%, n=5), EDTA plasma (range 81%–99%, average 90%, n=5) and heparin plasma (range 80%–89%, average 84%, n=5). Kit components should be stored at +4°C or -20°C as instructed in the protocol and not used past the expiration date.
