From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) enables the in vitro quantitative determination of porcine Connective Tissue Growth Factor (CTGF) in serum, plasma, tissue homogenates, and other biological fluids. The kit is reactive with porcine samples and uses a colorimetric detection format read at 450 nm in a microplate reader. An antibody specific for CTGF is pre-coated onto a 96-well microplate; standards and samples are added, followed by a biotinylated anti-CTGF antibody and an HRP-avidin conjugate. TMB substrate produces a blue product that turns yellow upon addition of acidic stop solution, with the resulting absorbance proportional to CTGF concentration.

The assay has a sensitivity of <0.242 ng/ml and a working range of 0.625–40 ng/ml, with a total assay time of approximately 4 hours 30 minutes. Reported recovery across n=5 samples averages 92% for serum (range 82–102%), 97% for EDTA plasma (range 91–103%), and 93% for heparin plasma (range 86–101%). Linearity assessed across 1:2 to 1:16 dilutions yields serum recoveries of 81–101%, EDTA plasma 80–102%, and heparin plasma 79–104%.
