This sandwich ELISA from Antibodies.com is configured for the in vitro quantitative determination of porcine Cathepsin K in serum, plasma, or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) bearing an antibody specific for Cathepsin K. Standards and samples are added; after washing, a Biotinylated Anti-Cathepsin K Antibody followed by an HRP-Avidin conjugate are applied. TMB substrate generates a colour change read at 450 nm on a colourimetric microplate reader, with the Cathepsin K concentration in each sample calculated from a standard curve.

Reported performance includes an assay range of 0.312–20 ng/ml, a sensitivity of 0.112 ng/ml, and an assay time of 4h 30m. Spike-and-recovery averages for serum (n=5), EDTA plasma (n=5), and heparin plasma (n=5) are 91%, 90%, and 84% respectively. Dilution linearity across 1:2 to 1:16 is reported for each matrix. Kit components are a pre-coated 96 well microplate, lyophilized standard (2 vials), Detection Solution A (120 μl), Detection Solution B (120 µl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml), and plate sealers (5 adhesive strips).

Storage temperatures are component-specific: the plate, standard, Detection Solutions, and Sample Dilution Buffer are held at -20°C, while the Wash Buffer, TMB Substrate, and Stop Solution are held at +4°C. The manufacturer states the kit should not be used past its expiration date.
