Porcine cardiac Troponin I ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of porcine Cardiac Troponin I in serum, plasma, tissue homogenates, and other biological fluids. The assay is specific for Cardiac Troponin I, with no reported cross-reactivity with other analogs or variants.

Standards and samples are added to the wells of a pre-coated 96-well microplate arranged as 12 × 8 well strips, where Cardiac Troponin I binds to the immobilised capture antibody. Following incubation and washing, a biotinylated detection antibody is added to form an antibody–antigen–antibody sandwich complex. HRP-Streptavidin conjugate and TMB substrate are then used to visualise the reaction, producing a blue product that turns yellow upon addition of the acidic stop solution. The intensity of the yellow colour is read at 450 nm and is directly proportional to the concentration of Cardiac Troponin I, which is calculated by reference to the standard curve.

The assay has a working range of 31.25–2000 pg/mL and a sensitivity of 18.75 pg/mL, with the complete protocol taking approximately 4 hours. Serum and plasma samples should be diluted at least 1:2 with Sample Dilution Buffer prior to testing; validated dilutions of 1:2, 1:4 and 1:8 have been demonstrated for healthy serum as well as EDTA and heparin plasma. Kit components are shipped at +4 °C and stored at +4 °C or −20 °C as instructed in the protocol manual. Freeze/thaw cycles should be avoided and the kit must not be used past its expiration date.
