Description

This sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com is designed for the in vitro quantitative determination of porcine Anti-Müllerian Hormone (AMH) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay employs the sandwich enzyme immunoassay technique using a pre-coated microplate. An antibody specific for AMH is immobilised on the plate; standards and test samples are added and AMH present in each sample is bound to the wells by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-AMH Antibody is added, followed by an HRP-Avidin conjugate. After further incubation and washing, TMB substrate solution visualises the enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of an acidic stop solution. The density of yellow is proportional to the amount of AMH captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a colorimetric microplate reader and referring to the standard curve.

Key specifications include a measuring range of 0.156–10 ng/ml and a sensitivity of <0.041 ng/ml. The assay is run on a colorimetric pre-coated microplate platform with an assay time of 4h 30m. Recovery was validated in serum (range 80%–102%, average 91%, n=5), EDTA plasma (81%–99%, average 90%, n=5) and heparin plasma (80%–89%, average 84%, n=5). Kit components are to be stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.
