Description

Antibodies.com provides this Porcine ACE ELISA Kit, a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) intended for the in vitro quantitative determination of porcine Angiotensin I Converting Enzyme (ACE) in serum, plasma or other biological fluids. The assay is reactive with porcine samples and falls within the Assays and Kits product category.

The assay employs a pre-coated microplate to which an ACE-specific antibody has been immobilised. Standards and test samples are added to the wells and ACE present in each sample is captured by the immobilised antibody. After washing, a Biotinylated Anti-ACE Antibody binds the captured ACE, followed by addition of an HRP-Avidin conjugate and TMB substrate. The resulting blue-coloured product changes to yellow upon addition of acidic stop solution, with the density of yellow proportional to the amount of ACE captured. The concentration of ACE is calculated by reading the O.D. absorbance at 450 nm on a colorimetric microplate reader and referring to the standard curve.

Manufacturer-stated performance includes an assay range of 31.2–2000 pg/ml, a sensitivity of <10.7 pg/ml and a total assay time of 4h 30m. In spike-and-recovery testing, serum (n=5) returned values ranging from 80–102% with an average of 91%, EDTA plasma (n=5) gave 81–99% (average 90%) and heparin plasma (n=5) gave 80–89% (average 84%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions was: serum (n=5) 82–96%, 83–98%, 81–99%, 93–101%; EDTA plasma (n=5) 88–101%, 86–95%, 90–102%, 80–93%; heparin plasma (n=5) 80–91%, 82–90%, 95–104%, 79–95%. Kit components should be stored at +4°C or -20°C as instructed and the kit must not be used past its expiration date.
