Porcine Alpha-1-Acid Glycoprotein ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is intended for the in vitro quantitative determination of porcine alpha 1 Acid Glycoprotein (AGP) in serum, plasma, or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, using an antibody specific for alpha 1 Acid Glycoprotein/AGP that has been pre-coated onto a microtiter plate.

Standards and test samples are added to the wells, where the target analyte is captured by the immobilised antibody. Following incubation and washing, a biotinylated anti-AGP antibody is added, followed by an HRP-Avidin conjugate. After further incubation and washing, TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The intensity of the yellow colour, read at 450 nm in a microplate reader, is proportional to the amount of AGP captured in each well, and the concentration can be calculated by reference to a standard curve.

The kit has a sensitivity of <0.19 ng/ml and an assay range of 0.39-25 ng/ml. Detection is colorimetric on a pre-coated microplate in a 12 x 8 well strip format, with a total assay time of 4h 30m. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date. Validation data showed average recovery in serum of 80%-102% (n=5), in EDTA plasma of 81%-100% (n=5), and in heparin plasma of 80%-89% (n=5), with dilution linearity demonstrated at 1:2, 1:4, 1:8, and 1:16 across the same sample types.
