Description

From Antibodies.com, this is a sandwich Enzyme-Linked Immunosorbent Assay intended for the in vitro quantitative determination of mouse Ku70 (X-Ray Repair Cross Complementing 6) in tissue homogenates or other biological fluids. The assay uses a 96-well microplate pre-coated with an antibody specific for Ku70. Standards and samples are added to the wells, the analyte is captured by the immobilised antibody, and detection is achieved with a Biotinylated Anti-Ku70 Antibody followed by HRP-Avidin conjugate. TMB substrate is then added, producing a blue product that turns yellow on addition of the acidic stop solution; the yellow intensity is proportional to the amount of Ku70 captured in each well, and the concentration is calculated from the O.D. absorbance read at 450 nm against the standard curve.

The quantitative detection range is 0.312–20 ng/ml with a sensitivity of <0.107 ng/ml, read on a colourimetric microplate reader using the pre-coated 12 × 8-well strip plate. The complete assay time is 4 h 30 min.

Manufacturer validation (n=5 per matrix) reports mean recoveries of 91% in serum (range 80–102%), 90% in EDTA plasma (range 81–100%) and 84% in heparin plasma (range 80–89%). Dilution linearity has been demonstrated at 1:2, 1:4, 1:8 and 1:16 across all three matrices. Kit components should be stored at +4 °C or −20 °C as instructed, and the kit must not be used past its expiration date.
