A sandwich ELISA from Antibodies.com is designed for the in vitro quantitative determination of mouse WFS1 (Wolfram Syndrome Protein 1) in tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate configured as 12 x 8 well strips. WFS1 in standards or test samples is captured by an immobilised antibody, detected with a Biotinylated Anti-WFS1 Antibody, and visualised using an HRP-Avidin conjugate and TMB substrate; the enzymatic reaction produces a blue colour that changes to yellow on addition of the acidic stop solution. Absorbance is read at 450 nm on a microplate reader and the concentration of WFS1 is calculated against the standard curve.

The quantitative assay range is 0.312–20 ng/ml with a sensitivity of <0.133 ng/ml, detection is colourimetric, and the full protocol takes 4h 30m. Kit components are stored at +4°C or –20°C as instructed, and the assay must not be used past the kit's expiration date.

Spike-and-recovery performance has been characterised across serum, EDTA plasma and heparin plasma (n=5 each), with average recoveries of 91% (range 80–102%) for serum, 90% (range 81–100%) for EDTA plasma and 84% (range 80–89%) for heparin plasma. Dilution linearity has been assessed at 1:2, 1:4, 1:8 and 1:16, with reported ranges of 87–91%, 87–107%, 74–101% and 92–97% for serum, 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma, and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (n=5 each).
