This sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com is designed for the in vitro quantitative determination of mouse Von Willebrand Factor A Domain Containing Protein 1 in plasma. The assay uses the sandwich enzyme immunoassay technique on a pre-coated microplate of 12 x 8 well strips. The manufacturer refers to the target as Warp. Standards and samples are added to the wells, where the target is captured by an immobilised specific antibody. After incubation and washing, a Biotinylated Anti-Warp Antibody binds the captured target, an HRP-Avidin conjugate is added, and TMB substrate is then used to visualise the enzymatic reaction. The reaction produces a blue colour that changes to yellow on addition of acidic stop solution, and absorbance is read at 450 nm in a microplate reader. Total assay time is 4 hours 30 minutes.

Performance specifications include a sensitivity of <12.4 pg/ml and an assay range of 31.2-2000 pg/ml. Sample recovery (n=5) in serum was 80%-102% (average 91%), in EDTA plasma was 81%-100% (average 90%), and in heparin plasma was 80%-89% (average 84%). Dilutional linearity has been reported at 1:2, 1:4, 1:8, and 1:16 dilutions across serum, EDTA plasma, and heparin plasma samples. The assay is read on a colorimetric microplate reader and is suitable for plasma.

Kit components comprise a pre-coated 96 well microplate, lyophilised standard (2 vials), Detection Solution A (120 µl), Detection Solution B (120 µl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml), and plate sealers (5 adhesive strips). Components are stored at +4°C or -20°C as instructed and should not be used past the indicated expiration date.
