The kit employs a pre-coated 96-well microplate arranged as 12 x 8 well strips, together with a biotinylated anti-c-Myc detection antibody, an HRP-avidin conjugate, and a TMB substrate solution. The enzymatic reaction is read colorimetrically at 450 nm using a standard microplate reader, with the intensity of the yellow product proportional to the amount of c-Myc captured in each well.

With a total assay time of 4 hours and 30 minutes, the kit offers a sensitivity of less than 0.056 ng/ml and a quantitative range of 0.156–10 ng/ml. Recovery has been demonstrated in serum (range 80–102%, average 91%, n=5), EDTA plasma (range 81–100%, average 90%, n=5), and heparin plasma (range 80–89%, average 84%, n=5).
