Following incubation and washing, a Biotinylated Anti-TIE1 Antibody is added, which binds the captured TIE1 in each well. An HRP-Avidin conjugate is then added, and after further incubation and washing, TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution.
