The assay employs a sandwich enzyme immunoassay format on a pre-coated 96-well microplate arranged as 12 x 8 well strips, with absorbance read on a colorimetric microplate reader at 450 nm.

Reported sensitivity is <0.101 ng/ml with a working range of 0.312–20 ng/ml, and the complete protocol takes approximately 4 hours 30 minutes. A BCMA-specific capture antibody is pre-coated on the plate; standards and samples are added, followed by a Biotinylated Anti-BCMA Antibody and an HRP-Avidin conjugate. TMB substrate is then added, producing a blue product that turns yellow upon addition of stop solution; the intensity of yellow colour is proportional to BCMA concentration, which is calculated against a standard curve. Kit components include a pre-coated microplate, lyophilized standard (2 vials), Detection Solution A (120 μl), Detection Solution B (120 μl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml), and plate sealers.
