Description

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of mouse alpha Tubulin in tissue homogenates, cell lysates or other biological fluids. The assay uses a pre-coated microplate platform arranged as 12 x 8 well strips and is read on a colorimetric microplate reader at 450 nm.

The quantitative measurement is based on a sandwich enzyme immunoassay technique: an antibody specific for alpha Tubulin is pre-coated onto the microplate, standards and test samples are added into the wells and bound, the wells are washed, and a Biotinylated Anti-alpha Tubulin Antibody together with an HRP-Avidin conjugate is used for detection. TMB substrate solution visualises the HRP enzymatic reaction and the colour changes to yellow upon addition of acidic stop solution; the density of yellow is proportional to the amount of alpha Tubulin captured. The reported assay range is 0.312-20 ng/ml with a sensitivity of <0.114 ng/ml, and the total assay time is 4h 30m. Recovery averages (n=5) were 91% for serum (range 83%-100%), 95% for EDTA plasma (range 91%-99%) and 94% for Heparin plasma (range 90%-99%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 was reported as 81-110%, 82-98%, 90-100% and 80-108% for serum (n=5); 91-97%, 90-107%, 70-108% and 89-93% for EDTA plasma (n=5); and 86-104%, 79-103%, 95-99% and 86-110% for Heparin plasma (n=5).

Components supplied include a pre-coated 96 well microplate (12 x 8 well strips), lyophilized standard, Detection Solution A, Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.
