Mouse Tryptase ELISA Kit

Supplied by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay is intended for the in vitro quantitative determination of mouse Mast Cell Tryptase in serum, plasma, tissue homogenates or other biological fluids. The assay uses a pre-coated 96-well microplate configured as 12 x 8 well strips. A capture antibody specific for the target is immobilised on the plate; standards and samples are added, and any Mast Cell Tryptase present is bound by the immobilised antibody. After washing, a biotinylated anti-Mast Cell Tryptase detection antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow on addition of acidic stop solution. The intensity of yellow is proportional to the amount of Mast Cell Tryptase captured in each well and is read at 450 nm on a colorimetric microplate reader.

The quoted assay range is 78.1-5000 pg/ml and the sensitivity is below 31 pg/ml. The total assay time is approximately 4 hours 30 minutes. Recovery has been reported in serum (range 80-102%, average 91%), EDTA plasma (range 81-100%, average 90%) and heparin plasma (range 80-89%, average 84%). Dilution linearity has been evaluated at 1:2, 1:4, 1:8 and 1:16 across the same matrices. Kit components are stored at +4°C or -20°C as instructed in the protocol, and the kit must not be used past its expiration date.
