The Antibodies.com Mouse Trypsinogen Activation Peptide (TAP) ELISA Kit is a competitive enzyme immunoassay that quantitatively measures the short N-terminal activation peptide cleaved from trypsinogen during its conversion to active trypsin. Because TAP is liberated only upon proteolytic activation of the zymogen, it serves as a direct biomarker of premature intrapancreatic trypsinogen activation, which is the initiating event in acute pancreatitis and a contributor to pancreatic injury in cystic fibrosis, duct-obstruction, and drug-induced pancreatic toxicity models.

The 96-well microplate is pre-coated with TAP antigen, and mouse serum, plasma, or pancreatic tissue homogenate samples are co-incubated with a mouse-reactive anti-TAP antibody, so that free peptide in the specimen competes with the immobilized antigen for binding. After washing, a chromogenic TMB substrate is added and read at 450 nm, with TAP concentration inversely proportional to absorbance. The kit is supplied as a complete set including pre-coated strip wells, mouse TAP standards, detection antibody, substrate, stop solution, and wash buffer, sufficient for one full standard curve and replicate sample wells in a single run. A recommended 50 µL sample volume per well is used for both standards and unknowns.

Storage and handling: unopened kit components are kept refrigerated at 2-8 degrees C, the lyophilized standards are reconstituted and aliquoted for single-use storage, and the chromogenic substrate should be protected from light. Reconstituted reagents remain stable for the period indicated on the lot-specific certificate of analysis when handled under these conditions. Researchers using caerulein hyperstimulation, L-arginine, or choline-deficient ethionine-supplemented diets to induce experimental pancreatitis in mouse models typically pair this competitive ELISA with amylase and lipase activity assays, pancreatic histology, and trypsin activity readouts to fully characterize the zymogen-activation cascade.
