Mouse Toll Like Receptor 7 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com enables the in vitro quantitative determination of mouse TLR7 in tissue homogenates, cell lysates or other biological fluids. The kit uses a 96-well microplate pre-coated with an antibody specific for TLR7. Standards and samples are added to the wells, and any TLR7 present is captured by the immobilised antibody. After washing, a biotinylated anti-TLR7 antibody binds the captured TLR7, followed by addition of an HRP-avidin conjugate. A TMB substrate is then used to catalyse an enzymatic reaction producing a blue product that changes to yellow upon addition of acidic stop solution.

The density of yellow colour is proportional to the amount of TLR7 captured per well, with concentration calculated by reading the absorbance at 450 nm in a colorimetric microplate reader and referring to the standard curve. Assay time is 4 hours 30 minutes. Sensitivity is 0.061 ng/ml, with a product range of 0.156–10 ng/ml. Reactivity is Mouse. Kit components include a pre-coated 96-well microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and adhesive plate sealers. Components should be stored at +4°C or -20°C as instructed and the kit must not be used past its expiration date.

Recovery was evaluated across five replicates per sample type. Serum yielded an average recovery of 91% across a range of 80%–102%; EDTA plasma gave an average of 90% (range 81%–100%); heparin plasma gave an average of 84% (range 80%–89%).
