Mouse Toll Like Receptor 4 ELISA Kit

Designed for the in vitro quantitative determination of mouse Toll Like Receptor 4 (TLR4), this sandwich ELISA from Antibodies.com is suitable for tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated microplate (12 x 8 well strips) on which an antibody specific for TLR4 has been immobilised. Standards and samples are added to the wells; captured TLR4 is detected using a Biotinylated Anti-TLR4 Antibody followed by an HRP-Avidin conjugate. TMB substrate produces a blue colour that turns yellow on addition of acidic stop solution, and absorbance is read at 450 nm in a colorimetric microplate reader, with TLR4 concentration calculated from the standard curve.

Reported characteristics include a product range of 0.156–10 ng/ml and a sensitivity of <0.065 ng/ml. Sample recovery has been evaluated in serum (80–102%, average 91%, n=5), EDTA plasma (81–100%, average 90%, n=5) and heparin plasma (80–89%, average 84%, n=5). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 was reported for serum (87–91%, 87–107%, 74–101%, 92–97%), EDTA plasma (90–105%, 84–101%, 90–101%, 79–108%) and heparin plasma (84–95%, 92–105%, 82–105%, 89–91%). Total assay time is 4 h 30 m, and kit components are stored at +4°C or -20°C as instructed.
