A sandwich enzyme-linked immunosorbent assay (ELISA) from Antibodies.com, designed for the in vitro quantitative determination of mouse TIMP2 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique: an antibody specific for TIMP2 is pre-coated onto a 96-well microplate, standards and test samples are added into the wells and the TIMP2 present in each sample is bound by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-TIMP2 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate produces a blue-coloured product that changes to yellow after the addition of acidic stop solution, with absorbance read at 450 nm in a colorimetric microplate reader and TIMP2 concentration calculated by reference to the standard curve.

The assay has a standard range of 0.312–20 ng/ml and a sensitivity of <0.131 ng/ml, with a total assay time of 4 h 30 min. Undiluted sample recovery averaged 91% in serum (range 80–102%), 90% in EDTA plasma (range 81–100%) and 84% in heparin plasma (range 80–89%), each across n=5. Dilution linearity at 1:2, 1:4, 1:8 and 1:16 dilutions gave recoveries of 87–91%, 87–107%, 74–101% and 92–97% for serum (n=5); 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma (n=5); and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (n=5). Kit components are stored at +4°C or −20°C as instructed and the kit must not be used past its expiration date.
