Description

Antibodies.com supplies a sandwich Enzyme-Linked Immunosorbent Assay (ELISA) kit for the in vitro quantitative determination of mouse TARC/CCL17, also known as Thymus Activation Regulated Chemokine, in serum, plasma or other biological fluids. The kit is intended for use with mouse samples and the assay is read on a colorimetric microplate reader.

The kit uses the sandwich enzyme immunoassay technique. An antibody specific for TARC/CCL17 is pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells, and TARC/CCL17 present in each sample is captured by the immobilized antibody. After washing, a Biotinylated Anti-TARC/CCL17 Antibody binds the captured analyte, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The intensity of the yellow colour is proportional to the amount of TARC/CCL17 captured, and concentration is calculated by reading the O.D. absorbance at 450 nm against the standard curve.

The assay range is 31.2–2000 pg/ml with a sensitivity of 11.3 pg/ml, and the complete protocol runs in approximately 4 hours 30 minutes. Reported recovery in undiluted serum was 80%–102% (average 91%, n=5), in EDTA plasma 81%–100% (average 90%, n=5), and in heparin plasma 80%–89% (average 84%, n=5). Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
