Mouse Thymidine Phosphorylase ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of mouse Thymidine Phosphorylase in serum, plasma, or other biological fluids. The kit employs a pre-coated 96-well microtiter plate (12 x 8 well strips) with an immobilised capture antibody specific for Thymidine Phosphorylase. Standards and samples are added to the wells, followed by a Biotinylated Anti-Thymidine Phosphorylase Antibody and HRP-Avidin conjugate. After washing, TMB substrate is added and the reaction is stopped with acidic stop solution; the intensity of the yellow colour is proportional to the amount of Thymidine Phosphorylase captured. Absorbance is read at 450 nm in a colorimetric microplate reader and the concentration is calculated against the standard curve.

Reported assay sensitivity is <0.142 ng/ml with a measuring range of 0.312–20 ng/ml. The total assay time is 4 h 30 m. According to the manufacturer, recovery in serum ranged from 80%–102% (average 91%, n=5), in EDTA plasma 81%–99% (average 90%, n=5), and in heparin plasma 80%–89% (average 84%, n=5). Kit components should be stored at +4°C or −20°C as instructed, and the kit must not be used past its expiration date. The kit is classified under CLP for skin irritation, eye irritation and skin sensitisation, and is intended for laboratory/research use only.
