Mouse Thrombospondin 2 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of mouse Thrombospondin 2 in plasma, tissue homogenates or other biological fluids. The assay uses an antibody specific for Thrombospondin 2 pre-coated onto a 96-well microplate. Standards and samples are added to the wells, where Thrombospondin 2 is captured by the immobilised antibody. After washing, a Biotinylated Anti-Thrombospondin 2 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then introduced; catalysis produces a blue product that turns yellow upon addition of acidic stop solution, with the density of the yellow colour proportional to the amount of Thrombospondin 2 captured in each well.

Concentration is calculated from the optical density absorbance at 450 nm read on a colorimetric microplate reader, by reference to a standard curve. Sensitivity is <0.124 ng/ml and the assay range is 0.312–20 ng/ml. The typical total assay time is 4 h 30 min. Reported recovery values are: serum (n=5) 80–102% (average 91%), EDTA plasma (n=5) 81–100% (average 90%) and heparin plasma (n=5) 80–89% (average 84%). The product is intended for plasma and tissue homogenates; kit components are stored at +4°C or −20°C as instructed in the protocol manual, and the kit should not be used past its expiration date.
