Mouse Syntaxin 1A, Brain ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) supplied by Antibodies.com for the in vitro quantitative determination of mouse Syntaxin 1a in tissue homogenates or other biological fluids. The assay uses the sandwich enzyme immunoassay technique, in which standards and test samples are added to wells pre-coated with an antibody specific for Syntaxin 1a. After incubation and washing, a Biotinylated Anti-Syntaxin 1a Antibody is added, followed by an HRP-Avidin conjugate and TMB substrate. Addition of acidic stop solution produces a yellow colour whose intensity is proportional to the Syntaxin 1a concentration, calculated by reading the absorbance at 450 nm against a standard curve.

The kit has a sensitivity of <0.057 ng/ml and a measuring range of 0.156–10 ng/ml, with a total assay time of 4 hours 30 minutes. The pre-coated microplate is supplied as 12 x 8-well strips and detection is performed colorimetrically using a standard microplate reader. Kit components are stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date. In validation testing, average recovery values were 91% in serum (n=5, range 83%–100%), 96% in EDTA plasma (n=5, range 90%–103%) and 95% in heparin plasma (n=5, range 89%–102%). Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16 (n=5 per sample type). For serum, observed ranges across the four dilutions were 83%–110%, 79%–100%, 90%–98% and 82%–108%. For EDTA plasma, ranges were 90%–98%, 90%–105%, 73%–109% and 87%–91%. For heparin plasma, ranges were 88%–103%, 77%–105%, 95%–98% and 87%–110%.

The kit is supplied with a pre-coated 96-well microplate, lyophilised standard, detection solutions A and B, wash buffer (30X), sample dilution buffer, TMB substrate, stop solution and plate sealers.
