Mouse Syndecan 1 ELISA Kit

Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of mouse Syndecan 1 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay employs a pre-coated microplate platform and a colorimetric detection method read at 450 nm in a microplate reader. Standards and samples are added to wells pre-coated with an antibody specific for Syndecan 1; following incubation, wells are washed and incubated with a Biotinylated Anti-Syndecan 1 Antibody, then with HRP-Avidin conjugate. TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow after the addition of acidic stop solution. The density of yellow is proportional to the amount of Syndecan 1 captured in each well, and concentration is calculated by referring absorbance readings to a standard curve.

The quantitative range is 125-8000 pg/ml with a sensitivity of 47 pg/ml, and the total assay time is 4h 30m. Validation in serum (n=5) showed average recovery of 91% (range 80%-102%), in EDTA plasma (n=5) 90% (range 81%-100%), and in Heparin plasma (n=5) 84% (range 80%-89%). Dilution linearity was demonstrated across 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and Heparin plasma. Kit components are stored at +4°C or -20°C as instructed and the components must not be used past the stated expiration date.
