The procedure employs a microtitre plate pre-coated with an antibody specific for SDF1; standards and test samples are added, followed by a biotinylated anti-SDF1 detection antibody and an HRP-avidin conjugate. A TMB substrate solution develops a blue product that turns yellow upon addition of acidic stop solution, with absorbance read at 450nm on a colorimetric microplate reader.

The quantitative assay covers a working range of 0.156-10 ng/ml with a sensitivity of 0.059 ng/ml and a total run time of 4 hours 30 minutes. Reported recovery values (n=5) are 80%-102% (average 91%) for serum, 81%-100% (average 90%) for EDTA plasma and 80%-89% (average 84%) for heparin plasma, with linearity data reported across 1:2, 1:4, 1:8 and 1:16 sample dilutions.
