Description

From Antibodies.com, this Mouse STAT3 ELISA Kit is a sandwich enzyme-linked immunosorbent assay (sELISA) configured for the in vitro quantitative measurement of mouse Signal Transducer and Activator of Transcription 3 (STAT3) in tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay is performed on a pre-coated 96-well microplate arranged as 12 x 8 well strips and read colorimetrically at 450 nm using a standard microplate reader, with a total assay time of 4 hours 30 minutes.

The kit applies the sandwich enzyme immunoassay principle. An antibody specific for STAT3 is pre-coated onto the microplate wells; standards and test samples are added and STAT3 present in each sample binds to the immobilised antibody. After washing, a Biotinylated Anti-STAT3 Antibody binds the captured STAT3, followed by an HRP-Avidin conjugate. TMB substrate is then added; the HRP-catalysed reaction yields a blue product that turns yellow on addition of the acidic stop solution. The intensity of yellow is proportional to the amount of STAT3 captured in each well, and the concentration is calculated from the 450 nm absorbance read against the standard curve. Kit components should be stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.

Reported performance characteristics include a measuring range of 0.781-50 ng/ml and a sensitivity of <0.31 ng/ml. Spike-and-recovery testing (n = 5 per matrix) returned average recoveries of 91% for serum (range 80-102%), 90% for EDTA plasma (range 81-100%) and 84% for heparin plasma (range 80-89%). Dilution linearity is also reported for serum, EDTA plasma and heparin plasma at 1:2, 1:4, 1:8 and 1:16 dilutions, with recovery ranges varying by matrix and dilution factor.
