Mouse Sestrin 2 ELISA Kit

Antibodies.com supplies a sandwich enzyme-linked immunosorbent assay (sELISA) for the in vitro quantitative determination of mouse SESN2/Sestrin-2 in tissue homogenates or other biological fluids. An antibody specific for SESN2/Sestrin-2 is pre-coated onto a 96-well microplate. Standards and samples are added, the target is captured by the immobilized antibody, and bound SESN2/Sestrin-2 is detected using a Biotinylated Anti-SESN2/Sestrin-2 Antibody followed by an HRP-Avidin conjugate and TMB substrate. The enzymatic reaction yields a blue product that turns yellow after addition of acidic stop solution; absorbance is read at 450 nm in a colorimetric microplate reader and concentration is calculated from the standard curve.

According to the manufacturer, the assay has a product range of 0.156–10 ng/ml and a sensitivity of 0.049 ng/ml. The total assay time is 4 hours 30 minutes. Reported spiking recovery ranges (n=5) were 80–102% (average 91%) for serum, 81–100% (average 90%) for EDTA plasma and 80–89% (average 84%) for heparin plasma, with dilution linearity data provided at 1:2, 1:4, 1:8 and 1:16 for each sample type. Kit components include a lyophilized standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, stored at +4°C or –20°C as instructed.
