Mouse Sestrin 1 ELISA Kit

From Antibodies.com, this sandwich ELISA is designed for the in vitro quantitative determination of mouse SESN1 (Sestrin 1) in tissue homogenates or other biological fluids. The assay employs a pre-coated 96-well microplate (12 x 8 well strips) onto which an antibody specific for SESN1 has been immobilised. Standards and samples are added to the wells, followed by a biotinylated anti-SESN1 detection antibody and an HRP-Avidin conjugate. A TMB substrate is then used to visualise the enzymatic reaction, producing a blue colour that turns yellow upon addition of the stop solution; absorbance is read at 450 nm on a colorimetric microplate reader and the SESN1 concentration is calculated by reference to a standard curve.

The kit reports a sensitivity of <0.041 ng/ml and a quantitative assay range of 0.156–10 ng/ml, with a total assay time of approximately 4 hours 30 minutes. Spike-and-recovery has been characterised in serum (range 80–102%, average 91%), EDTA plasma (range 81–100%, average 90%), and heparin plasma (range 80–89%, average 84%). Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.
